4-Dimethylaminopyridine is a novel and highly efficient catalyst that has been extensively employed in chemical synthesis in recent years. The electron-donating dimethylamino group in its structure resonates with the parent ring (pyridine ring), strongly activating the nitrogen atom on the ring for nucleophilic substitution. It significantly catalyzes the acylation (phosphorylation, sulfonylation, carbonylation) reactions of highly sterically hindered and low-reactive alcohols and amines, with an activity approximately 10^4 to 10^6 times that of pyridine. It exhibits high catalytic capability in various types of reactions, including acylation, alkylation, etherification, esterification, and transesterification, in organic synthesis, pharmaceutical synthesis, pesticides, medicine, dyes, fragrances, polymer chemistry, and analytical chemistry. It has an extremely noticeable effect on improving yield. Additionally, it can be used as a phase transfer catalyst for interfacial reactions. Its advantages include: low catalyst usage, typically only 0.01–5% of the substrate molar amount; the acid produced in the reaction can be neutralized with organic or inorganic bases; mild reaction conditions, generally proceeding at room temperature, saving energy; a wide range of solvent choices, applicable in both polar and non-polar organic solvents; short reaction time, completing reactions in minutes with DMAP compared to prolonged reactions with pyridine, thus greatly enhancing labor productivity; high yields, achieving 80–90% for sterically hindered hydroxyl compounds that hardly react with pyridine, thereby improving reaction yield and product quality while simplifying the process; few side reactions, low odor, and minimal waste generation. Due to its excellent catalytic performance, DMAP is referred to as a "super catalyst" and has become one of the most commonly used catalysts for organic synthesis practitioners. In the domestic chemical pharmaceutical industry, it has been successfully applied in the production of active pharmaceutical ingredients such as acetyl(spiramycin), artemisinin succinate, and statin lipid-lowering drugs, improving process conditions and yielding good economic and social benefits. Furthermore, it is widely used in the total (semi) synthesis of complex natural products, such as: acetylation in the total synthesis of PRAVASTATIN, trifluoroacetylation in the total synthesis of TUBERCIDIN, acetylation in the total synthesis of MUGINEIC ACID, benzoylation in the total synthesis of TERPESTACIN, and acylation in the total synthesis of Sildenafil (VIAGRA). In pesticide production, the use of DMAP in the synthesis of tetramethrin has improved yield and product purity. It also shows significant catalytic activity in the synthesis of carbamates from isocyanates and in the reaction of chrysanthemoyl chloride to synthesize pyrethroids. Its role is quite notable in the synthesis of organophosphorus compounds through phosphorylation reactions.
