Description: Dextran sulfate is a polyanionic derivative of dextran, formed by the esterification reaction of dextran and sulfuric acid. It contains approximately 17% sulfur, equivalent to an average of 1.9 sulfate groups per glucose residue in the dextran molecule.

Key Applications:

·Enhances nucleic acid hybridization rate - A solution containing 10% dextran sulfate increases the reannealing rate of DNA strands by approximately 10-fold. This phenomenon further enhances the hybridization rate of single-stranded or double-stranded probes with membrane-bound DNA/RNA. Moreover, the addition of 10% dextran sulfate may increase the hybridization rate of randomly cleaved double-stranded DNA probes with immobilized nucleic acids by up to 100-fold.

·Exhibits immunologically related activities:

(1) Enhancement and suppression of humoral immunity;

(2) Polyclonal activation of B lymphocytes, even stimulating immature B cells;

(3) In thymocyte responses, promotes transformation into agglutinins;

(4) Inhibits blood coagulation and platelet aggregation;

(5) Enhances fibrinolytic activity;

(6) Enhances/suppresses cell-mediated immune responses;

·Precipitates low-density (LDL) and very low-density (VLDL) lipoproteins - In the presence of Mg²⁺, dextran sulfate precipitates low-density lipoproteins from serum, leaving high-density lipoproteins in the supernatant. This lipoprotein precipitation function of dextran sulfate can also be used to purify other materials, such as β-2-glycoprotein.

·Acts as an adjuvant;

Solubility:

Soluble in water (100 mg/ml - clear or slightly hazy yellow solution; high molecular weight products are less soluble in water than low molecular weight ones). Aqueous solutions should be buffered (e.g., with sodium carbonate) before sterilization to prevent degradation.
