The viable mycoplasma are lysed, releasing enzymes that catalyze a reaction with the substrate provided in the kit, converting ADP to ATP. By measuring the ATP levels before and after adding the substrate, a ratio is obtained that indicates the presence of mycoplasma. If these enzymes are absent, there is no increase in the second reading compared to the first; if the enzymes react specifically with the substrate, it results in an increase in ATP levels. The rise in ATP is detected through a bioluminescent chemical reaction, as per the following formula: LuciferaseATP + Luciferin + O2 ——————> Oxyluciferin + AMPMg 2+ + PPi + CO2 + LIGHTThis reaction occurs at room temperature (18°C to 22°C), which is the optimal temperature for the luciferase reaction. In the detection principle of the mycoplasma detection kit, the final spectrophotometric reading shows that the intensity of light emitted is linearly related to the concentration of ATP, allowing for the detection of mycoplasma contamination. |
