Application of Chromatographic Separation Technology in the Separation and Purification of Natural Products:
In the separation and purification of traditional Chinese medicine, commonly used chromatographic techniques include adsorption chromatography and ion-exchange chromatography. Adsorption chromatography encompasses reverse-phase chromatography (C18 reverse-phase filler) and normal-phase silica gel chromatography. Ion-exchange chromatography is primarily used for the separation and purification of charged compounds. Currently, chromatographic separation technology plays an increasingly important role in refining active ingredients of traditional Chinese medicine, such as flavonoids, alkaloids, glycosides, organic acids, and polysaccharides.
During percolation, the solvent penetrates the cells of the medicinal material, dissolving a large amount of soluble substances. As the concentration increases and the density rises, the solvent moves downward. The upper extraction solvent or dilute extract replaces its position, creating a favorable concentration gradient that allows diffusion to occur naturally. Thus, percolation offers better extraction efficiency than maceration, achieves more complete extraction, and saves time by eliminating the need for separating the extract.
