The separation system mainly consists of the chromatographic column, which plays a key role in separation. When using chromatographic columns, especially polar ones, it is essential to remove oxygen from the carrier gas. Care should be taken to avoid introducing air when replacing gas cylinders, and a helium purifier can also be installed in the gas flow path.

If issues such as ghost peaks occur, consider trimming a portion of the column length at the inlet end first. Depending on the baseline and column bleed, the column should be conditioned in a timely manner. Column conditioning generally follows the principle of slow heating and fast cooling: start at 50 °C, increase at 5 °C/min to 20 °C below the upper temperature limit, hold for 30 min, then decrease at 20 °C/min to 50 °C. Repeat this cycle 2-3 times. Note that the column should be disconnected from the detector during conditioning to avoid contaminating the detector. If non-volatile residues in the column still cannot be removed, trim 30-50 cm of the column at the inlet end and recondition the column.

When the chromatographic column is no longer in use, both ends should be sealed with discarded septa to isolate the column from external air, preventing air ingress and damage to the internal coating.
